Abstract: Previous studies have demonstrated that bovine chromaffin cells cultured in medium with 10 nM insulin-like growth factor-I (IGF-I) secrete about twofold more catecholamine when exposed to secretory stimuli than do cells cultured without IGF-I. The purpose of this study was to determine whether protein kinase C (PKC) is involved in the effect of IGF-I on secretion from these cells. PKC was down-regulated in the cells by 16–18 h of treatment with β-phorbol didecanoate (β-PDD; 100 nM). Such treatment had no effect on high-K+-stimulated secretion from cells cultured without IGF-I; however, secretion from cells cultured with IGF-I was reduced to a level comparable to that in cells cultured without the peptide. The inactive isomer, α-PDD (100 nM), had no effect on secretion from untreated or IGF-I-treated chromaffin cells. The effect of β-PDD was time and concentration dependent, with 100 nM β-PDD producing a maximal effect in 8–10 h. In situ PKC activity measured in permeabilized cells treated with PMA (300 nM) was decreased by~40% by 10 h and was reduced to almost basal levels by 18 h. Immunoblotting experiments demonstrated that both α-and ε-PKC were lost from the cells with time courses similar to that seen in the in situ PKC assay. Overnight treatment with the PKC inhibitor H7 (100 μM) prevented the enhanced secretion normally seen in IGF-l-treated cells, whereas HA1004 had no effect. High-K+-stimulated 45Ca2+ uptake in IGF-I-treated cells was attenuated by long-term treatment with β-PDD (200 nM) or H7 (100 μM). Together these observations suggest that PKC is required for IGF-I-enhanced secretion from chromaffin cells.  相似文献

A leucine/valine substitution at amino acid position 127 was identified by the polymerase chain reaction and restriction fragment length polymorphism in the bovine growth hormone gene. Genotyping was performed in 84 AI bulls of three different breeds, in which plasma concentrations of growth hormone (GH) and insulin-like growth factor I (IGF-1) were also measured. Gene frequencies of variants L (leucine) und V (valine) were 0.80/0.20 (Black and White), 0.90/0.10 (Brown), 0.71/0.29 (Simmental). Hormone concentrations were measured during different physiological conditions (normal feeding, fasting, realimentation) in the majority of animals. Generally, genotype LL was associated with higher concentrations of GH than LV. This difference was significant in Black and White bulls (P < 0.05). In contrast, IGF-1 concentrations were higher in LV than in LL animals. This was most pronounced in mature, realimented Simmental bulls. We conclude that the various GH alleles influence the circulating concentrations of GH and IGF-1.  相似文献

Abstract A number of bacteria were isolated from different anoxic reactors. Those having denitrifying potential were tested for their ability to denitrify under aerobic conditions. The activity of their denitrifying enzymes varied from partial inactivation by oxygen (strains NO2B9 and TCET1) to oxygen-independent activity in a strain named SGLY2 which was tentatively identified as Comamonas sp. The effect of different aeration conditions on growth and on denitrification of SGLY2 was studied more extensively. This strain was able to consume oxygen and nitrate simultaneously with the production of nitrogen and without build-up of nitrite. The dissimilatory nitrate-reductase of nitrate-adapted cells was found to be more active in the presence of oxygen than in micro-aerobic or strictly anaerobic conditions.  相似文献

Resprout and mature plant shoot growth, leaf water status and gas exchange behavior, tissue nutrient content, flowering, and production were studied for co-occurring shallow-rooted (Arbutus unedo L.) and deeprooted (Quercus ilex L.) Mediterranean tree species at the Collserola Natural Park in Northeast Spain Resprouts showed higher growth rates than mature plant shoots. During fall, no differences in eco-physiological performance of leaves were found, but mobilization of carbohydrates from burls strongly stimulated growth of fall resprouts compared to spring resprouts, despite low exposed leaf area of the fall shoots. During summer drought, resprouts exhibited improved water status and carbon fixation compared to mature plant shoots. Shoot growth of Q. ilex was apparently extended due to deep rooting so that initial slower growth during spring and early summer as compared to A. unedo was compensated. Tissue nutrient contents varied only slightly and are postulated to be of minor importance in controlling rate of shoot growth, perhaps due to the relatively fertile soil of the site. Fall flowering appeared to inhibit fall shoot growth in A. unedo, but did not occur in Q. ilex. The results demonstrate that comparative examinations utilizing vegetation elements with differing morphological and physiological adaptations can be used to analyze relatively complex phenomena related to resprouting behavior. The studies provide an important multi-dimensional background framework for further studies of resprouting in the European Mediterranean region.  相似文献

Abstract The cellular fatty acid composition of the psychrophilic Vibrio sp. strain No. 5710 isolated from a deep-sea sediment sample was analyzed. The presence of docosahexaenoic acid (22:6) was demonstrated as found previously in other deep-sea bacteria, and the relative amount of 22:6 decreased as the growth temperature increased. A temperature shift from 10°C to 0°C resulted in a relative increase of 22:6, and an opposite shift led to a decrease. In addition, hexadecanoic acid (16:0) was found to increase as the growth temperature increased. Therefore, it is suggested that the adaptation of 5710 to the growth temperature was carried out by the changes in the relative amounts of 22:6 and 16:0. When 5710 was grown at low temperature, it increased the relative amount of 22:6 presumably to maintain membrane fluidity at that temperature. In contrast, 5710 grown at high temperature probably maintained the membrane fluidity by increasing the amount of a saturated fatty acid, 16:0. Furthermore, observation of the fatty acid compositions at mid-exponential phase and early stationary phase revealed the proportions of several fatty acids, including a major fatty acid, 9- cis -hexadecenoic acid (16:1c, palmitoleic acid), were affected by the growth phase which may be due to the physiological difference between the growth phases.  相似文献

An important component of the interaction between macroinvertebrates and leaf litter in streams in the extent to which consumers can differentiate between undecomposed and decomposing leaves. The detritivores Gammarus pulex and Asellus aquaticus fed preferentially on conditioned rather on unconditioned leaf material. Growth in A. aquaticus was significantly reduced when unconditioned leaves were provided, but in G. pulex no significant effect of conditioning on growth was observed. The capacity of G. pulex to tolerate reductions in food quality seems to be a consequence of a compensatory system in which respiration rates change to compensate for reductions in food quality. In this way a constant growth rate is maintained. Increases in ingestion rates to compensate for low quality food were not observed.  相似文献

Rainbow trout were used to characterize the direct influence of growth hormone on hepatic lipid mobilization in vitro. Liver was removed from fish fasted 72 h, sliced into 1-mm3 pieces and incubated in Hanks' medium containing ovine or salmon growth hormone (0.28 ng·ml-1–28 g·ml-1). Lipid mobilization, resulting from triacylglycerol hydrolysis, was evaluated by fatty acid and glycerol release into culture medium. Control rates of fatty acid release and glycerol release were 0.95±0.16 (mean ± SE) and 0.88±0.28 mol·l-1·mg fresh weight, respectively. Both ovine growth hormone (28 ng·ml-1) and salmon growth hormone (28 ng·ml-1) stimulated fatty acid release into culture medium, increasing rates by 112% and 97%, respectively, during the course of a 24-h incubation. Glycerol release was similarly augmented by growth hormone treatment. Growth hormone-stimulated metabolite release became evident within 12 h and persisted for up to 72 h. The presence of amino acids in the culture medium potentiated the lipolytic action of growth hormone. Ovine growth hormone (28 ng·ml-1) in the presence of amino acids stimulated a 53% increase in fatty acid, and a 108% increase in glycerol, release over rates observed in the absence of amino acids. Salmon growth hormone (28 ng·ml-1) in the presence of amino acids stimulated a 53% increase in fatty acid, and a 44% increase in glycerol, release over rates observed in the absence of amino acids. Ovine growth hormone (28 ng·ml-1) also stimulated gluconeogenesis, as indicated by a 75% increase in phosphoenolpyruvate carboxykinase activity in liver pieces incubated in the presence of amino acids. The effects of a high-growth genetic background on the growth of mice hemizygous for one of two growth hormone transgenes were examined. Male mice hemizygous for wild-type (W) and dwarf mutant (M) bovine growth hormone (bGH) transgenes were crossed with females of a high-growth selected (S) and control (C) line as follows: W x S, W x C, M x S and M x C. Body weights of progeny were recorded weekly from 2 to 10 weeks of age. F1 progeny were classified as carriers (P) or non-carriers (N) of the transgene by assaying tail DNA for bGH using the polymerase chain reaction and agarose gel electrophoresis. A deficiency in the number of f1 progeny carrying the W (P<0.05) and M (P<0.01) bGH transgene was most likely due to differential prenatal and early postnatal mortality. Bodyweight means of wild-type transgenic mice were larger (P < 0.05) than those of non-transgenic littermates by 3 weeks of age in a C background in contrast to 5 weeks in S. The wild-type bGH transgene increased adult body weights more in the C (155%) than in the S (136%) background, indicating transgene expression by selection background interaction (P < 0.05). However, the growth response to the wild-type transgene in the S background was still large. The dwarf mutant transgene had a greater effect on growth reduction in the S (70%) than in the C (84%) background, thus causing transgene expression by selection background interaction (P < 0.05). Gender by wild-type transgene effect interactions (P < 0.001) for adult body weight were caused by the transgene reducing the gender difference for body weight in C and eliminating it in S. The dwarf mutant caused a larger negative effect on growth in males than in females, resulting in a gender by dwarf mutant transgene interaction (P < 0.001) for adult body weights. Results indicate that the effect of a GH transgene on growth can be affected both by a high-growth genetic background and the gender of progeny.  相似文献

Arnica montana, one of the character species of the replacement plant communityViolion caninae on sandy acid humic podzol, is declining in the Netherlands since 1950. Locally, it is even extinct.This process of decline may be attributed to (i) autonomic succession; (ii) increased rate of acidification of the soil and/or (iii) change in competitive relations amongArnica and more productive plant species, such as the successive dominantDeschampsia flexuosa. This paper examines the last two hypothesized factors, supposedly being regulated by atmospheric input of N-input, by (a) describing the habitat characteristics of a seemingly still healthy individual-rich population ofArnica and (b) growingArnica andDeschampsia in mixed humic podzol soil (mainly A1 horizon) fertilized with variously-composed nutrient solutions in order to assess the nutrient supplying capacity of the podzol soil and species-specific nutritional demands related to their respective growth capabilities.The results suggest that an increased rate of soil acidification yielding extra supply of ionic Al and Mn is presumably of less importance. The implications of growth rate differences amongArnica andDeschampsia as related to their nutritional demands are likely far more important.Arnica grows more slowly thanDeschampsia in fertilized humus podzol. The latter species has a much more efficient use of nutrients. Furthermore, both species differ in K-nutrition if NH4 + is the dominant N-source, a situation normally occurring in acid podzolic soils.Deschampsia possesses the capability to adapt its metabolic nutrition (avoiding nutritional imbalances) to a wide range of nutrient supplies thereby maintaining a rather constant level of growth.Arnica, on the contrary, lacks this capability. These results are discussed in the framework of competitive relations among co-occurring plant species in the plant communityViolion caninae. It is concluded that maintenance ofArnica and likely other character species of that alliance — all characterized by a low growth rate — will only be achieved when the plant community is properly managed by trampling, mowing or light levels of grazing (low stocking rate). Of prime importance is to maintain a low and relatively open vegetation structure at a relatively low level of nutrient supply.  相似文献
